311. CLOSTRIDIUM MEDIUM I

K₂HPO₄ 3.0 g

KH₂PO₄ 2.0 g

NH₄Cl 2.0 g

MgCl₂ × 6 H₂O 0.2 g

CaCl₂ × 2 H₂O 0.05 g

Yeast extract 5.0 g

Na-resazurin solution (0.1% w/v) 0.5 ml

D-Glucose 5.0 g

L-Cysteine-HCl × H₂O 0.5 g

Na₂S × 9 H₂O 0.5 g

Distilled water 1000.0 ml

Dissolve ingredients except glucose, cysteine and sulfide. Sparge medium with 100% N₂ gas for 30–45 min to make it anoxic, then dispense under same gas atmosphere into anoxic Hungate-type tubes or serum vials and autoclave. Add glucose, cysteine and sulfide from sterile anoxic stock solutions prepared under 100% N₂ gas. Adjust pH of the complete medium to 7.0-7.2, if necessary.
