Glucose 1.0 g

KH₂PO₄ 0.2 g

NaCl 0.2 g

NH₄NO₃ 0.2 g

MgSO₄ × 5 H₂O 0.04 g

Yeast extract(without sodium chloride) 0.2 g

Vitamin solution (see below) 1 ml

Distilled water 1000.0 ml

pH 5.0-6.5

Vitamin solution:

p-Aminobenzoic acid 1.0 mg

Biotin 0.2 mg

Nicotinic acid 2.0 mg

Thiamine-HCl 1.0 mg

D-Ca-pantothenate 0.5 mg

Pyridoxamine 5.0 mg

Vitamin B₁₂ 2.0 mg

Distilled water 1000.0 ml

Prepare the medium without the vitamin solution. Autoclave at 121°C for 15 min and add the vitamin solution from a filter-sterilised stock solution.
