Solution 1:

MgSO₄ x 7 H₂O 0.5 g

NH₄Cl 0.5 g

KH₂PO₄ 0.5 g

CaCl₂ x 2 H₂O 0.05 g

KCl 0.025 g

Ammonium acetate 0.5 g

Ferric citrate solution 5.0 ml

Trace element solution SL6 (see below) 2.0 ml

Vitamine B₁₂ 20 μg

Distilled water 1000.0 ml

Solution 2 (Ferric citrate solution):

Ferric citrate 0.1g

Distilled water 100 ml

Solution 3 (Trace element solution SL-6):

ZnSO₄ x 7 H₂O 0.1 g

MnCI₂ x 4 H₂O 0.03 g

H₃BO₃ 0.3 g

CoCl₂ x 6 H₂O 0.2 g

CuCl₂ x 2 H₂O 0.01 g

NiCl₂ x 6 H₂O 0.02 g

Na₂MoO₄ x 2 H₂O 0.03 g

Distilled water 1000.0 ml

Solution 4:

NaHCO₃ 10.0 %

Solution 5:

Na₂S × 9 H₂O 10.0 %

Solution 6:

Na₂S₂O₃ 10.0 %

All solutions are prepared aerobically and autoclaved at 112°C for 30 minutes. Before inoculation solutions 4, 5 and 6 are added to the medium (solution 1) with final concentration of 2 g per 1 liter for NaHCO₃ and (if necessary) Na₂S₂O₃ and 1 g per 1 liter of medium for Na₂S x 9 H₂O. The pH of the final medium adjust to 7.0-7.3.
